高野山の地主神を祀る丹生都比売神社（天野社）の４つの祭神を描く。空海が高野山に金剛峯寺を開創した際、その土地を譲った神および道案内をした神として「丹生都比売明神」（丹生明神）「狩場明神」（高野明神）が登場する。この両明神が、真言密教の護法神として勧請された。さらに、承元２年〈1208〉には、平政子が敦賀国（福井県）の気比神社から大食都比売神（気比明神）、安芸国（広島県）の厳島神社から市杵嶋比売神（厳島明神）を勧請合祀した。ここに高野四所明神が成立した。高野四所明神像は、本殿に、四神それぞれが桐樹を描いた金地屏を背にして牀座に坐す神像の形式に描かれる。右上の第１殿丹生明神と第３殿の気比明神は唐装束の女神像、第２殿狩場明神は束帯姿の男神像、第４殿の厳島明神は琵琶を弾く童女像の姿に描かれる。欄干には一対の狛犬、階段下には白黒の犬を配している。現存する高野四所明神像は、室町時代以降のものに限られる。金泥・截金で繊細で華麗に描写された本図は、その作風から室町時代後期・16世紀の制作と思われる。
